1999年在《失魂少女》中，墨菲饰演一位精神分裂的病患，与薇诺娜·瑞德、安吉丽娜·朱丽一同飙戏而崭露头角，也让大家认识了这位外型甜美中带着一丝不羁的女孩；2001年由迈克尔·道格拉斯主演的惊悚片《一言不发》特别钦点布莱特妮·梅菲担任女主角伊莉莎白，剧中她饰演18岁的重度精神病患者，抢眼的表现令人印象深刻。知名杂志《名利场》特别在封面专题，点名她为好莱坞下一波最受期待的女星之一。2002年与阿姆合演广受好评的《8英里》，在其中扮演阿姆的女友，2003年又与好莱坞新晋红小生阿斯顿·库彻联手主演了喜剧片《新婚告急》，她天真浪漫又风骚性感的演出，又让她赢得年轻版美国甜心的封号。
